While I am not accustomed to the breadth of wealth Dr T had back then, I feel 400k at the initial investment for something classed as speculative would still be significant. I guess its a very subjective point, as you've stated.
I think the importance that Dr T didn't sell any shares to fund his options is much more telling of where he expects bisantrene to sit in comparison to other investment choices he may have.
Maybe we'll know how much wealth he has when/if he exercises his remaining options and he doesn't sell any heads to fund them
