@IndexInvestor there is the definite possibility that there may be a synergy between an FTO inhibitor and an anti-CD47 drug.

The oncogene MYC regulates the expression of CD47 (and PDL1) (1). Suppression of MYC in mouse and human tumor samples led to a reduction in the expression of CD47 (and PDL1). MYC bound diredtly to the promoters of CD47 (and PDL1). Inactivation of MYC down-regulated the expression of CD47 (and PDL1) and enhanced the anti-tumor immune response. The City of Hope 1.0 paper found that through inhibiting FTO, Zantrene was able to reduce the expression levels of MYC (2; figure below). If we are to connect these dots, then an FTO inhibitor could very well improve the efficacy of an anti-CD47 drug by decreasing the expression levels of CD47 through downregulation of MYC (as well as exerting it's own anti-tumor effects on sensitivity, metabolism, immune evasion, growth and proliferation, cell cycle arrest, and apoptosis signalling).

Of course, there are other articles that suggest inhibition of knockdown of FTO decreases the expression level of MYC in other cancer types (3, 4, 5, 6).
