MOZZ NOTES - PART 1
Yeah this is going to be a labour of love
All below is my own opinions and cannot be misconstrued as advice.
Looks like (Thanks OX) the official poster is out...this means I can finally post on it.
It will still take me a full 2 days to sift through my notes and articulate what I want to say...so bare with me but I'll release it in parts.
Tonight we cover Part 1 - The Poster.
So as we can see there is some divergence but the good news here is that it is GOOD DATA!
The interesting thing is that there is somewhat divergent data...look at that table above...1 week trumped (was better) than 2 week dosing!
HUH?
Soooooo what did Womac do, was that the same?
NOPE!
The above Womac was for the TWO week dosing regime.
So this is the anomaly. Its very interesting...let me summarise to make it bleedlingly obvious
WET Biomarkers (TnfA, Args etc) were better compared to 1 week dosing
Womac Observations did better with 2 a week dosing.
How is this possible...
I have asked a lot of ppl this...I mean a lot, and not just randoms.
The expert opinions so far can be summarised as follows
1) The synovial readings is a SNAPSHOT
This is a very important point. Anchor Man asked me about this. I thought for a sec and came back with this analogy.
In the old days we used to have film...this was actually a frame by frame production and when you run through them quickly enough, you get moving images....lovely...
A Horse running...frame by frame, put it together and she runs...
So the synovial markers is a single frame, a single snapshot at a point in time...it might show the horse with a leg forward..it might show the leg at some other position...it might be a family video and the third cousin on the right actually has a sad face in a given instance, a given frame...but a different instance she smiles and she looks a lot more pretty and a lot more happy...
It COULD be the same here... at that point of time, that's how it was.
Take the snapshot a month earlier and the two weekly dosing could've been a whole lot different...yeah, take it at the 6 month point (coming to a thread near you) and it may or may not be the same?
2) Imagine this scenario..the two weekly dosed guys get such lovely pain relief they think dang it man...I AM going to hit that gym...I am going to go for that long awaited walk by the ocean...and in doing that they load up on the biomechanical stress and perhaps there is some influence on the wet markers in the synovium? Ie brings them down in the two weekly dosing cohort more so than the one week.
Lets, finally, GO!
3) This one is a proud Mozzie hypothesis...I ran this by one of the world's best earlier this morning at Oarsi when I had her all to myself....(I cant say any more, don't ask me)
We all know the bell curve, let me whip it out for you one more time....
Imagine the peak is our 2 m/kg. Now imagine a lovely balanced ONCE a week dosing regime...what happens? The drug is injected, the body's processes it over the week...and we get the magic happening...yeah yeah we can get all sciency processes but now is not the time....
BUT contrast this with TWICE a week dosing regime for instance, that's more loading., that's less time to clear...that in effect COULD be more cumulative dosing and thus we are moving the needle (sorry) a little more to the right on that above bell chart....this means that we are losing some efficacy...thus the two week synovial observations drop compared to the 1 week dosing?
Only a hypothesis, but I got a nod and a "you could be right" by the world's BEST.
That was enough for me..the money I spent on the trip was prob validated by me meeting and having a real one on one.
I have So so much more to say ....just bare with me...
One last very important point to close off this part 1....
We do really need to wait for our 6 month data, have patience...
oh and dont forget...DYOR
