Caught up with RH about slide24 of the presentation.
The following is my interpretation that hopefully can be understood:

The proceedure Phylogica use to determine if their 'functional penetrating peptides(FPP) and cargo' can successfully enter the inner-cell, involves the use of a fluorescent protein(GFP).

A protein fragment(S11) is removed from GFP and linked to Phylogica's FPP/cargo.
    With S11 removed, GFP(now GFP1-10) stops glowing.

In the assays the GFP1-10 protein is inserted into the cells, but not all cells accept the GFP1-10 protein. The measure of cell acceptance is termed the 'transfection efficiency'. So if the transfection efficiency is 70%(as on slide24), it means the GFP1-10 protein has entered 70 of every 100 cells.

If when tested, the protein combination FPP/cargo/S11 can pass through the cell membrane and endosome into the inner cell successfully, S11 will reunite with GFP1-10 to create a glowing GFP protein.

This glowing protein shows which cells have been efficiently entered by FPP/cargo.

When determining the success rate of cell entry by FPP/cargo, check the graph on slide24. It shows (PYC-FPP3_RBD_S11) on about 45%.

It must be remembered that if only 70% of the cells contained the GFP1-10 protein, then this will affect the result. Simple maths: (45/70)x100=64%.

The success rate of cell entry for (PYC-FPP3_RBD_S11) is round 64%.
