THE (PATIENT) INTERVIEW - PART 2In Part 1 we were introduced to Patient S. We got a sense of what a dire situation he was in. Luckily for him he was already a share holder and knew of this potential alternative called iPPS.
Part 2 continues with his personal account. Will his situation improve further? What were his detailed observations and indeed, what is all this about Mozz Bonuses?
Injection 7 - Bloods taken. Knees are very good now. Woke up today with very little pain! Knee a little sore again on next day morning but it eased up again by the end of the day... Now only one spot in knee feels pain, just some tightness remains.
Injection 8 - Bloods taken, knee has pain in one area but still feels tight. Pain overall around the 3/10 now.
Injection 9 - Pain in knee doesn't seem to be improving any further.
Injection 10 - Same sort of pain (around 3/10), mentioned to the prescribing Dr - but Dr suggests that many patients notice a drop in pain 3 to 4 weeks after the last injection.Two injections left to go!
Mozz additional notes: At the end of week 4, S. just had his 7th injection and on some days, while the pain varies a little, on some days and at certain times he is recording....ZERO pain....sometimes a 2 out of 10.
Paradigmers....Mr S now sleeps at night...pain free!
He still has 2 injections to go, I've also heard a number of patients experiencing less pain reduction even mid way or towards the end of the initial treatment BUT after a full month after the last injection they report much reduced levels of pain.
Its only one patient's detailed account above...but as I have said, I have heard a number of personal stories and we can also see the results in a much broader community of SAS patients let alone our clinical trials.

THE MOZZ BONUSOk alright, what's all this talk about a Bonus?
Disclaimer: Now please don't think I'm saying iPPS will cure every indication there is ... please don't think I'm ever daring to say we will cure or even treat each and every case of inflammation and OA and any single aliment.
What I will say is that without it being advice, in my humble personal opinion, I think we could positively affect many....yeah quite a few future patients we may just have on our books.
I also must state, I'm compelled to say that while I do oftentimes (all the time) sound enthusiastic...there are risks, some real risks that this play may not work out exactly how I want...exactly how YOU want....bare this in mind. NEITHER of us should be blamed if it doesn't...I see what I see...you do too...its up to you. ALWAYS ALWAYS work out where you will stand...(where will you sit) if this doesn't work out. Never invest in one and only one investment no matter how good it sounds. Speak to a licensed investment adviser if you need assistance.
A Mozz Bonus doesn't often happen, I've had like 3 of them in the past 4 years...tonight....its special...tonight we have a Mozz Bonus...
Mr S is a very happy man...he now has almost no pain, he still has 2 more shots left to go....he is sleeping at night and he has function back.
He told me he deliberately isn't going crazy with the exercise so that his body can just recover.
But as a Mozz Surprise Bonus I have not ONE but TWO pearlers...

Remember Mr S had borderline reportable blood pressure before the program of iPPS?
The nurse was on the verge of having to escalate it...Prior reading was... 140/90...that's borderline too high (see Appendix B)
Here's what I deliberately left out from the diary entries above by week...what I didn't show you...This is how the results went at a couple of time points throughout Mr S's course:
Week 3 - Blood Pressure down to 140/80, nurse is happy
Week 4 - Blood pressure coming down slowly
Week 5 -Blood pressure now 125/85, great result.

NORMALISEDWe know that there are three major areas of the body where our magic juice is predominantly absorbed, Kidney, Liver and Spleen. Its the kidney's that, amongst a number of vital functions, it is charged with the responsibility of regulating blood pressure. No doubt when you are in pain there is a chance of higher blood pressure, but definitely there is some pathophysiological processes taking place that has a direct positive effect here. As I mentioned, there is another one...another Mozz bonus tonight and this one I have personally NEVER come across before...but before the reveal, we do need some background...
THE PROSTRATEThe prostrate gland's most important function is the production of a fluid that, together with sperm cells from the testicles and fluids from other glands, makes up semen.

(I have none of those staff above..sure, one day I will have my own office and a decent editor! Yeah I'm dreaming).
Hold my calls dear secretary...a Mozz scoop is in the making!...but unfortunately I'm not the first to this sorta observation...yeah there were a bunch of researchers that got on to this recently...ok well not that recently..

20 years ago someone knew that ol' PPS has some effect on the prostrate?
Here is the paper written 20 years to the day (well the day when I was researching for this post):
I'll give you a few more details on the high level contents of this paper in Appendix D below.
Their conclusion was quite a read.

Nope, its not ONLY because he is just sleeping fully through the night DUE TO LACK OR LESS pain...... there is another separate reason...one pertinent to us....
Its because of another AMAZING phenomena that's going on that I couldn't guess...that I didn't know about...that I discovered after a very insightful chat to Mr S.

Mr S's prostrate was slightly enlarged BEFORE iPPS, this was confirmed by a Doctor that did some diagnostics on this circa a year ago... After the first 7 or so iPPS injections.....MR S is not having to get up to go to the toilet as much, "usually either once max. or not at all" (in his exact words)...It appears that iPPS has reduced the inflammation around this gland/muscle....this would have the direct effect of reducing pressure against his bladder...this alleviates the necessity of having to go to the toilet so often.OH MY GOODNESSThis is what you own.

STOP PRESSJust before going to print I got an update from Mr S. He just had his 11th injection and he is at a 2 out of 10 pain now (another step down from last week's base line of a 3/10)...The best and latest line I got from him just yesterday?
"Mozz, I'm feeling a lot happier"
The additional thing I love about Paradigm (apart from the super stellar product we have exclusively). is the fact that management aren't just aiming to acquire a pass into the biggest markets in the world. Yeah they are wanting to smash all records totally. They are aiming so high that they will make our own

They are not only wanting to sew up the OA market and other indications, but they are also preparing and planning AFTER market...ie after arthroplasties, ie Post Op Pain. We have patents in place.
How many patients are still in pain after surgery?
"...although joint arthroplasty offers good pain relief and improves quality of life, up to 20% of patients complain of persistent pain".7Its been found that this persistent pain in some of the post op patients may not be resolved even after a full year after the surgery and in these cases NSAIDS and or corticosteroids are often prescribed up to or instead of revision surgery.
"Revision surgery is complicated, comes with the risk that pain will not diminish, and in some cases can be detrimental".7
"However, non-steroidal anti-inflammatory drugs (NSAIDs) and corticosteroids have been found to have negative effects on bone healing and the metabolism of cartilage). The risk of fracture, both traumatic and spontaneous, is increased in those subjects that take continuous corticosteroid therapy".7Paradigmers, PAR have patented the use of iPPS in the case of post op pain. This is the company I want to invest in. A company that does not merely aim for a 40% increase....or a X2...nah they are thinking real long term and they are planning to be front line best in class in an indication that gets measured in the Multi Billions and spans across many sub variants in the Arthritic domain.
Not to mention all the other inflammatory type future indications.

Now remember in our recent announcement we received this observation of TIMPS?

Well again we see that these researchers also noted a similar finding.
5"Increase in inhibitor of cartilage degrading enzymes TIMP-1".The next extract from the research shows us just HOW much of an effect is being sighted. Zymography
9 is a process of observing enzyme's actions in a gelatin substance. Typically it is used to observe the actions of some of the MMP's.
"Tissue inhibitor of MMP (TIMPS), examined by reverse zymography, increased 200% over control".These consistent and key findings all add to the background of our drug, the way in which it works and the most amazing observations that are being uncovered. Paradigmers, its not just one independent research team reporting the effects of our molecule. Its consistent findings AND its prolific and material.
"The amount of αI type (IV) and αI type (I) collagen released in the supernatant, measured by ELISA, significantly decreased in PPS-treated cultures".MMP-9 is one of the most widely investigated MMPs. It is responsible for regulating the remodelling processes of inflammation and fibrosis in diseases such as cardiovascular disease.
10MMP-9 is a good protein to have. But why bring this up here?
Because the paper said this about it:
"MMP-9 activity increased 234.4% over control". What P value for that observation? P was <0.001.
Here is another quote from another set of researchers looking at MMP-9:
"MMP-9 cell-specific overexpression, however, has also proven beneficial and highlights the fact that little information is available on the underlying mechanisms of MMP-9 function". ...and further,
"MMP-9 plays important roles in immune cell function".Lets see some of this data in chart form:
Filters play a vital role in many domains...there are filters on our taps, in our cars...there are filters within our air conditioners and yes, even we humans have them. They play such an important multi role that you could argue that's why we have not one, but two of these units. Its the kidneys that amongst a number of things like filtration, also regulate the blood pressure.
The research into Pentosan's miraculous and efficacious effect is not brand new.
It was all the way back in 2010 researchers observed the effects of Pentosan induced into rats specifically looking at Pentosan's action on the renal function.
11The title in itself is fairly illuminating:
"Pentosan polysulfate treatment preserves renal autoregulation in ANG II-infused hypertensive rats via normalization of P2X1 receptor activation".
There are a number of observations in the above paper, one such chart included here:
The conclusion states it utilising the words COMPELLING evidence and SIGNIFICANT renal protection.
"The results of this study provide compelling evidence that anti-inflammatory intervention with PPS confers significant renal protection against the decline of renal autoregulation and this is associated with improved reactivity to P2X1 receptor stimulation in ANG II-infused hypertensive rats".Incredible.
