Multi-billion dollar platform drugs almost always harmonize with SoC. Replacing it is very rarely accomplished because the clinical and commercial risk in doing so is absurdly high. As you might expect, there are few examples in the literature of replacing SoC, but there are numerous examples of drugs, like Keytruda, that found many homes next to SoC therapies.
Generally speaking, you need 2 pivotal phase 3 trials to prove that the new drug(s) is better than SoC. This is basic information that any reasonably well read biotech investor knows.
