For those that can't access it, this study uses benitec technology (shRNA) to knockdown two genes encoding proteins that suppress the activity of hypoxia-inducible factor (HIF), a master regulator of gene expression (+100 genes) switched on by heart cells starved of oxygen, as occurs with a myocardial infarct. HIF is critical to the heart cell survival mechanisms during the hypoxic episode, and regeneration post-injury.
The researchers induced a myocardial infarct in mice then, using benitec's patented technology (love it!), they enhanced the activity of HIF and were able to show some pretty remarkable outcomes, including a less damaged and better functioning heart, with improved blood supply to it. They also show how the enhanced HIF lead the restorative processes, by attracting more stem cells and inflammatory markers to the site of injury.
