The top part i.e. red line is the I-body. It has the basic scaffold of a human protein called n-CAM stitched with some parts of shark antibody to get the I-body. I-bodies have larger binding footprint I.e. it can bind targets that normal antibodies cannot bind. The bottom part shows the various ways they tried to increase their life in the blood. Since they are smaller than normal antibodies, they can get excreted through kidneys. They tried different methods and the best method is to make it a dimer or two molecules together by stitching with a portion of human antibody which is not involved in any binding (the Fc portion). That is shown on the right. That is the final product in trial i.e. ad214.
