Well you could look at it like but also he has a large amount of shares and options to execute, so he could actually benefit by stepping down to allow for a more commercial focused CEO to take over an lead the Co into the next stage. If he genuinely had some health issues and thought he wasn't the right person past the R&D stage it would be beneficial to his holdings and the Co as a whole for him to step down. A new commercial focused CEO will hopefully increase revenue and the SP so LDN would still be winning.
If he stepped down an proceeded to sell everything then I'd be a bit more concerned about the resignation personally
