Protease inhibitors are potent because they drop viral load by 4 logs on their own - yes telaprevir has rebound in monotherapy because resistance develops. This is why it needs interferon to maintain response. BIT225 only dropped the viral load 4 logs when given IN COMBINATION with interferon and riba - the drop attributable to BIT225 was only just under 1 log. In addition, ribavirin alone has no effect on viral load. It augments the effect of interferon and possibly other drugs through mechanisms unknown. It should be noted than an average baseline HCV viral load is 5 to 6 logs, so if PSI7977 reduced this to zero, even with ribavirin, it is extraordinarily potent, and at least 1000x moreso than BIT225. Yes it is in geno 2 and 3 which are relatively easy to treat with SOC, but it did this with ribavirin alone, a feat never before achieved, and a result that has changed the game. And of course interferon cause a lot of side effects (in HCV - strangely it is relatively benign in HBV), this is why SOC has to change.

    Worthind: nice searching - the quadruple therapy results are interesting, as previous non-responders are EXCEPTIONALLY difficult to treat, but as it is very difficult to get a majority of patients to accept just the PEG-IFN side effects, I think this regimen is only going to be applicable to a small percentage of highly motivated individuals, and will die out once the interferon-free protocols come on line, possibly as early as 2014.

    Rjaadave: you did exactly the right thing. Not necessarily because of what the share price might do, because many other factors effect that, but because you clearly shouldn't be in biotech shares. It's fine when the forum is full of uprampers who by and large have limited knowledge of the field, but when some valid questions are asked about the assumptions on the forum, your only response is to hurl abuse like some teenage schoolboy. Biotech is by its nature highly technical, so if you are investing you'd better have some idea about the technicals, or you'd be just as well spending your money on the slot machines at Crown Casino.
