I just took another peek into the PYC September update, and whilst the Phase 1 study is primarily to target safety, with patients receiving just one dose, with the dose volume dependant on which cohort they are enrolled into initially.

Further down the track,(later trials, Phase 2?), it makes sense that the focus will broaden to determine efficacy in humans which surely would include maintenance/repeat doses. Therefore the 75 microgram single dose cohort could possibly create some reference data which may provide insights into safety for when PYC get to the next step of multi dosing. And potentially the higher 75 microgram dose may also provide additional early clues for efficacy?

Shri definitely indicated that PYC were going to take up the fight/argument for an amendment for the Phase 1, (SAD) study to have a higher dose 4th cohort added. He mentioned 90 microgram at the time, however I am perfectly happy with Rohan's update pointing towards 75 microgram. I think we may be getting a fraction ahead of ourselves, because Shri mentioned the amendment would be sought after the 2nd cohort was completed, well we are not quite there yet... dosing completed, but the Safety Committee still needs to give the all clear for no adverse affects. This step is due fairly soon, and with that final piece we could possibly get the amendment being sought. (4th cohort for 75 microgram).

Don't forget folks, Shri's background before he joined PYC was that he was the head of Modality Innovation at the world's largest specialist Ophthalmology company, Stanton. Very impressive!!!
