Sure, lets stick to the facts.
ETOPS isn't irrelevant, particularly when flying across large oceans like the Indian ocean. The FAA may have approved an increase to 330 minutes (http://www.boeing.com/commercial/aeromagazine/articles/qtr_2_07/article_02_4.html), however this may be throttled-back (pun intended) if issues emerge.
As for aircraft configuration, QANTAS can reconfigure their A380s to carry many more passengers.
On the point of fuel consumption, I would like to see your numbers? Usually the higher the power rating, the more fuel is consumed. Four 787 power-plants may very well burn more fuel than four A380 power-plants. So show me the numbers please.
As for slots, most intelligent people would agree, that it is more economical and cost effective to fly in a single aircraft carrying more passengers than having to fly in two aircraft.
