What exactly are you concerned about with the City of Hope S2 trial?

There is no mention of FTO as an outcome measure anywhere in that link. If they are not measuring FTO and are purely looking at brequinars safety, tolerability, and efficacy in AML, then I think that they are comparitively a long way away from commercialisation than bisantrene. This is based on the massive amount of historic data of bisantrene in AML as well as more recent Sheba trial data that showed promising results.

Regarding IP associated with CoH findings of the mechanism of bisantrene. If you do an advanced search on patents, you will find Dr. T highlights that RAC hold the rights to bisantrene in many different forms in the US and around the world. He has also more recently discussed that it is very risky for a drug development team to use bisantrene, as patent law is very hard to disprove in drug development and the risk for CoH infringing on RACs patents would be quite high (link below). This is probably the reason why bisantrene was not enrolled into a follow up trial.

I'm not sure if you have heard about brequinar, but it failed as an anticancer agent in a trial with 250 patients as well as an organ transplant rejection drug. Also, brequinar was found to be impractical due to plasma concentrations varying 10-fold, making the dosing for each patient challenging and thus, impractical.

Opinion piece: Merchant have a trading strategy that they have stuck to, which indicates RAC are doing great. Garner held ~13 million shares, which he now still owns around 11.7 million. If I could sell down ~1.5 million shares to fund the next 3-5-10 years of my life, buy an Aston Martin, and live a life of simplistic luxury, while still holding >$40 million dollars worth of stock that, in all honesty, could go past being worth a $billion, I would too.
