It is not clear, however milestone increments in fee percentages are usually for amounts above milestones (eg. Sales $300M: fees $25M + $6M). The better payment is the milestone lump sum of $50M when sales achieve $250M, that is 25% of the milestone sales figure and is generous, but all lump sums are one-off.
There will be some misalignment with 'Annual Net Sales' and 'Net Sales in one calendar year' if it goes to the market in April, so the first lump sum won't be available until the end of 2024 now.
