I have written several posts in this thread as some sort of chronology of thoughts that may help with your series of excellent questions. I sympathise entirely in that I also fell asleep waiting for something positive to happen. Well its time to wake up for the opportunity is at hand.
You may have read all the posts or only the last post - I do not know. In the first post of the thread [Post #: 67992446] I discuss your very question of the liver sink effect that I believe threw up something of a red herring. In summary my understanding is that only the excess was distributed to the liver i.e. the drug reaches its target [high receptor occupancy] and works through IV administration.
AdAlta has followed a circuitous route but drug effect and the mechanism of action is often problematical and it takes time and process to reach the correct conclusions. I think we are there now which explains the push to re-commence clinical studies. The key question is who is doing the pushing apart of course from management. You may find my discussion on kidney fibrosis to be of interest in understanding the heightened sense of purpose.
