You'll see that, while mTOR complex 1 and 2 are 'distinct', they're also interrelated. The effect produced by up/down regulating signalling in either complex will depend on specifically where in the singalling process you're intervening, what cellular functions this intervention influences and any downstream or cascade effects.

Speaking very generally, it appears that MPL is influencing mTORC2 in a way that rapamycin (other rapalogues?) cannot or have not been able to thus far. I don't think the total affect of MPL on both mTORC1 and C2 has been fully described as yet.
