Actually, in this indication it is pretty linear.

Think of it this way. To treat sepsis caused by bacteria, the aim is to achieve a concentration of antibiotic in the blood which surpass the MIC (minimum inhibitory concentration) of the bacteria being targeted.

What is the blood volume of a rat (one of the safety study species)? ~65mL/kg liveweight.
What is the blood volume of a dog (the other safety study species)? ~80mL/kg liveweight
OK, what is the blood volume of a person? ~70mL/kg bodyweight.

So, relative to weight, blood volume is 7-8% regardless of whether your are a 300g rat or a 80kg man. So the same dose rate of antibiotic (in mg/kg) will achieve very similar blood concentrations.

Yes there would be differences in half-life due to species differences in excretion rates etc. But Recce has used the 50mg/kg figure as their anticipated effective dose for sepsis, with good reason. And from their material, it seems that higher doses may even be desired, particularly for other conditions.









Note: without PK/PD data such as half-life etc, it's hard to compare a 1-hour infusion with a 24-hour infusion. Maybe that's what you mean by non-linear@Bpharma ?
