40 samples was deemed by statisticians to be sufficient to provide statistical significance. A larger number wouldn't have made any significant difference to the results (statistically speaking).
Regarding people on placebo with decrease in Proteinuria vs baseline: If you read the results presentation, you will see that this is one of the areas the company is investigating further (and could lead to a reinterpretation of the significance of the overall results). It's called "legacy effect".
The results presented so far are "top line" results only, and further analysis of the results is likely to uncover additional insights.
