For a scenario where you're taking both pressures and samples you would try and achieve both on a single run but often hole conditions and tool configuration limitations necessitate two (or sometimes more) runs....first run to obtain pressures and second run to obtain samples. A formation testing tool with multiple probes and sampling kit attached is a very long tool and risk of getting the tool stuck is alot higher than for a shorter tool.As I've noted already you can only be sure that 1) you've successfully obtained a sample and 2) the liquid is what the telemetry tells you it is by checking the chambers once the tool is back at surface.
