When you have a drug used over multiple days and with cumulative effects then you can do a dose escalation by changing the number of days of treatment.
In this trial the maximum treatment time is only 5 days. It would be possible to modify the trial and add another dose escalation (say to 6 days) if there were too few dose limiting toxicities, but knowing what I know about these three drugs I strongly suspect 5 days of treatment is going to be close to the limit patients can handle.
