I think the only drug that doesn't include diarrhoea or constipation in the AE list is paracetamol... slight exaggeration there but hopefully my point is made. Retts patients all have constipation, are usually on aperients, so I'm skeptical if it is a true side effect. Could be more of a case of extremely effective bowel evacuation due to trofinetide on a background of constipation, which is often reported as diarrhoea, then the drug maker has no choice but to include it in the list of adverse effects. I think that would be common in this demographic. Solution, cease the aperients, add a tsp of metamucil or benefibre, continue trofinetide. IMO.
