So far BACE1 inhibitors have produced damage to the skin, eyes, and liver. The Merck one bothers me a bit because Merck has been accused of witholding negative side effects discovered in clinical trials (Vioxx and Fosamax, for instance). I believe that even if safe BACE1 inhibitors are developed they may only work at the very earliest stages of Alzheimer's disease and perhaps not even then.
