Playing the devil's advocate, I think if Dr kay was to respond to this letter he would say something along the lines of: "Criticise all you like but clinical trails show that AAV8 vectors are at least 20 times less effect in humans than they are in murine models. So how do you account for that?" See Dirk's blog.

So far I have not read any response that directly addresses this concern. The limited data from the haemophilia trials seems inconclusive to me.
