Chelator binding efficiency depends on the conditions and type/chemistry of the chelator. For example the pH (acid/base conditions) can be used to improve the efficiency of chelators binding to a radiometal, where for some chelators decreasing the pH (increasing acidity) can improve binding efficiency at room temperature, where those same chelators would require heating to say 90 degrees Celsius at more neutral pH conditions for efficient binding.

If the targeting ligand is sensitive to heat and low pH, e.g. antibodies, then clearly you want a chelator that bind efficiently to the radiometal under mild conditions. If it is a small molecule e.g. PSMA small molecule ligand, then heating and low pH might be ok. There are also other things to consider with the chelator, because modifying the targeting agent with a chelator can potentially change its biodistribution and targeting efficiency. Its a bit of a balancing act to achieve the best outcome.
