Good evening Optimus (Well Good morning actually)....Cartophen (I assume this is what you are referring to) does vary a little from animal to animal, take a read of this:
Cartrophen Vet has an optimal clinical efficacy in dogs at 3mg/kg of pentosan polysulfate sodium (PPS) administered sub-cutaneously on four occasions at intervals of 5-7 days. In the horse the recommended dosage is slightly lower at 2-2.5mg/kg of PPS administered by way of intramuscular injection on four occasions at intervals of 5-7 days. The dosage is equivalent to 1g of PPS per 400-500kg horse or the equivalent of a bottle of Cartrophen Vet (10mL) or Cartrophen Equine Forte (4mL), which contain 1g of PPS.
Ultimately for humans a 2x2 (2mg per Kg) over 6 weeks as you have mentioned seems to be close to the height of efficacy. Remember it is a Bell curve so too much will start to reduce in terms of efficacy.
At the same time we are all different so 2x2 is really an average.
Obviously, biologically there are differences between humans and various animals. We have certain biological aspects that are closer to some animals compared to others. So in other words it's not a simple translation to say that we are very similar to canines or we are very similar to Dolphins or Monkeys etc. It is going to depend on what biological aspect.
At any rate it has been determined after a number of years that 2x2 is pretty spot on. Of course you are going to get studies where certain individuals derived excellent efficacy with only 1 x 3 (as an example, this is a case study in regards to a patent) over just a few weeks....but at the end of the day it's been determined that 2x2 should give us a pretty good hit ratio.

PS: Don't forget, once we are licenced there will be a generally recommended dosage profile based on the Global harmonised Clinical Phase 3 we are undertaking and approved for. However, the Doc's will have some discretions to whether to allow their individual patients to go for a slightly longer dosage regime. Especially if this product is deemed to be quite safe. We kinda know it is with close to 1000 patients having tried iPPS and some 100 million injections in the past for various indications (DVT, IC etc).
