Yes, there is evidence of it potentially being harmful if too much is given, apparently I did read somewhere in the past that this dosage can be a fine line at a certain higher point. Eg 100 kg patient gets x mg max...if it was x + 30 mg its still ok..but x + 100 mg can be detrimental (all figures just an example).
Efficacy can be reduced at too low a dosage and too high a dosage.
Thus dosage segment as part of Phase III will determine best dosage. 2 mg/kg seems to be the current recommendation though.
