You are right that the 2a trial gave a lot more insight into how the drug works. Plasma results and MRI data will help scope out data for the 2b trial. This has also been used to look at expanding the indications it can be used for, and has given confidence to investigate combination studies.
They have pin pointed the bio marker that identifies where the drug will be effective. They will have screening for the trial that optimises the drug. Just like competitor drug companies do.
