However, currently available transcriptome-wide m⁶A-seq methods often need a large amount of RNA material (e.g., >20 μg total RNA), thus it is difficult to conduct m⁶A-seq for a large cohort of patients due to the limitation in primary patient samples. Therefore, much improved m⁶A-seq technologies that need much less RNA material and provide base-resolution m⁶A profiles are urgently needed. With such improved m⁶A-seq technologies, we can conduct m⁶A profiling with precious primary patient material or even with limited primary CSCs or TICs; the m⁶A profiles/signatures of certain particular transcripts or transcript loci could be identified as biomarkers for early cancer diagnosis, cancer classification, outcome prediction, and risk stratification. Besides cancer cells, cell-free circulating RNA (cfRNA) collected from plasma of cancer patients could also be used for m⁶A-seq, and particular m⁶A signatures could be used as biomarkers in clinic application.
