From my understanding PIIa studies are powered to find trends that can be explored in later stage trials and no necessarily Stat Sig. So the secondary endpoints of the PIIa are used as the primary in the PIIb/III where they will be setup to achieve. I'm not sure of the specific costs of the trials but I don't think the absolute number of people is necessarily in proportion to the cost. Some studies would be dirt cheap others could be very expensive
