They know how to interpret the pre-clinical trials and their optimism has been confirmed by the IND approval. There are different outcomes between pre-clinical and human clinical trials but IMU's experience must give them the confidence that those differences may be just minimal. That's why Prof. K and Prof Fong made those recent optimistic remarks in their videos.
