There's been some uncertainty of the thread about the significance of the GSK3 findings in the recent announcement.
The following may provide some clarity:
Of the 2 basic pathologies in AD (amyloid plaques and neurofibrillary tangles - NFT's) the role of PBT2 in amyloid patholgy is common knowledge on the forum. Little attention has thus far been paid to the NFT's.
NFT's are caused by the abnormal phosphorylation of Tau protein (which in its normal configuration helps provide rigidity and structure to transport channels within the neuron that carry nutrients and transmitters to the neuronal cell terminus).
Tau is phosphorylated by the GSK enzyme(a process that resultes in DE-phosphorylation of GSK.
If PBT2 in turn causes phosphorylation of GSK, the implication is that it is causing DEphosphorylation of Tau, and is thus putatively neuroprotective.
