So what do we know about PYC’s cell-penetrating peptide 1746 ??
1746 is one of PYC’s cell-penetrating peptides that they linked to the well researched omomyc protein drug molecule that inhibits the intracellular transcription factor MYC.
The linked molecule '1746-omomyc' was tested against a particularly aggressive form of breast cancer (MDA-MB-468) known to be hard to treat with conventional drugs.
At very low dosage signification cell death was observed (0.28um).
In the newsletter PYC write “To our knowledge this is the most potent direct inhibition of the MYC oncoprotein target yet described”.
Also in the newsletter they write “There is compelling genetic evidence that direct inhibition of MYC can lead to complete eradication of existing tumours”.
PYC have been building the research data to support their rhetoric.
So it appears 1746 is turning out to be a very special CPP.
PYC have also attached 1746 to other drug molecules. A peptide DPMI-a that targets MDM2 ubiquitin ligase and to a toxin Bouganin (which is only functional inside cells). With both DPMI-a and Bouganin intracellular delivery was enhanced.
So it’s possible 1746 could be linked to not only omomyc, but to other protein drug molecules that to-date have been unable to efficiently access inner cellular targets.
In the newsletter PYC write “Phylogica is now engaging with select academic and commercial collaborators to access a broader range of therapeutic cargoes, as well as their established cellular and animal model systems where we can rapidly evaluate functional activity”.
To write “we can rapidly evaluate functional activity” suggests PYC already have the cell-penetrating peptide ready to link to select customer’s protein drug molecules.
