Recent studies have identified mTOR inhibitors as promising therapies for epidermolysis bullosa simplex (EBS). Transcriptomic analyses revealed that mTOR inhibitors, such as sirolimus, can significantly oppose the gene expression signature associated with EBS blisters. Pilot studies using topical sirolimus showed marked clinical improvement in patients, reducing symptoms like painful plantar keratoderma. The mTOR pathway's involvement suggests that mTOR inhibitors could be effective in managing EBS by downregulating defective keratin expression and reducing cell proliferation, thus alleviating pain and itch.
Another prospective treatment, makes over 70 I believe.
