I’ve been giving a lot more thought to significance of Phylogica’s structurally diverse peptide libraries, imo - their core selling point.
Here’s what I understand...
Not only will this structural diversity apply to the Endosomal & Macrocyclic technologies, but also to the universal specificity Phylomer chip and numerous other applications of the newly patented synthetic phylomer libraries technology (once validated).
Do you recall the three D's highlighted in the Annual Report?
The enormous diversity of the Phylomer® libraries enables Phylogica to use them for three distinct purposes – referred to as the ‘three D’s’:
• To discover and validate new targets in phenotypic screens.
• To develop Phylomer blockers of these targets (often protein-protein interactions which are difficult to modulate by other means).
• To deliver Phylomer or protein cargoes to targets inside cells.
The ‘three D’s’ represents a unique combination of capabilities that clearly differentiates Phylogica from its competition.
For the newbies here, I have previously posted on the topic example I am highlighting below, but here’s a quick recap to illustrate just how significant ONE of these developments could be…
The ultimate goal of the collaboration is the construction of a universal specificity Phylomer chip. This low-cost consumable device will allow the identification of peptide probes that are specific for any target biomolecule, in a matter of minutes. This goal has not been achieved with existing approaches (eg. antibody arrays) on a single chip due to the intrinsic biases that are inherent with existing classes of biologics based on one structural family (eg. the antibody variable region). Even comprehensive arrays of antibodies, peptides of random amino acid composition, or protein scaffolds, cannot be used reliably to detect any biological antigen due to this issue of structural bias.

The reason I am so upbeat about this project is because the biosensor that Phylogica/IMB are working on will differentiate their product from anyone else in this space, as a result of the 'master keys' or structural diversity of their entire libraries. We are talking about truly disruptive technology here. This project is so under the radar at the moment and hardly any attention has been given to it, but do note, we are just on six months since the collaboration with IMB was announced to validate the Phylomer chip, so it shouldn’t take too much longer imo before we hear news/update on this front.
Whilst all focus is on Janssen news atm, let's not take our eyes off the numerous other projects in the mix. This may very well be one of those surprises that no one sees coming. :-)
