Targeting AP-1 Mediated Apoptosis for Skin Repair:

Cell death, particularly by apoptosis, is commonly activated by JNK-mediated phosphorylation of
c-Jun, a member of the Activator Protein-1 (AP-1) family of transcription factors. AP-1 proteins are
also involved in neuro-inflammation, tumorigenesis and other important cellular processes. After
a burn injury, the cells surrounding the injury site undergo cell death in a process similar to the
damage following a stroke or other neurotraumatic event. Glycinamide is a peptide inhibitor of
the AP-1 transcription factor, offering significant potential as a novel treatment for burns and as a
cosmeceutical for general skin repair.


AP-1 Inhibitor Peptides Discovery and Development:

Phylogica screened its Phylomer® library using an AP-1 regulated luciferase reporter gene assay to
identify a family of peptides which potently inhibit the AP-1 pathway. Phylogica enhanced the druglike
attributes of the peptides via TAT domain fusion, to improve the translocation of the peptide into
skin cells, and via retroinverso peptidomimetic modifications. Retro inversion involves replacement of
a D-isomer of an L-peptide with a reversal of the carbon peptide backbone, leading to decreased
peptide proteolysis and enhanced stability. These enhancements have led to attractive stability and
protease resistance.
Efficacy studies in models of full thickness burns were then used to select and validate a lead candidate,
PYC35B (Glycinamide). The results of these in vivo efficacy studies are summarised below.
