Antisense drugs have a pretty long history of liver toxicity.
Still not many drugs approved by the FDA despite many through clinic.
ISIS mipomersen or Kynamro with it's "blockbuster" $2B Genzyme deal and all the promise only gained a controversial approval and needed to come with boxed warnings with liver toxicity and low platelet counts - that ended up being a flop anyway with commercial uptake not delivering and they ended up selling it off for $100M. Spiranza also has much of the same type warnings attached ?
