Yes, and no.
A flow test will measure permeability over an extended time period, and therefore, over an extended area of the target reservoir.
It's the gold standard, nearly identical to actual production.
But you can take wellbore measurements, you can take core samples measurements, you can use specialist tools to isolate and flow test individual reservoirs.
The real question is, "How much accuracy, and how much data, do they want to pay for?".
