Perhaps if I can say it in short, notch (signaling) pathways play a major role in (tumour) cell proliferation through involvement in homeostasis regulation (stable conditions, e.g. PH level, temperature etc). If one can interrupt Notch pathways in a tumour cell, it will then have difficulty to survive or proliferate. Cell environment is very sensitive and often one needs to only slightly interfere with things homeostasis to end survival. Now, if Notch signaling can be inhibited, chemo drugs for example can more effectively "disturb" (the conditions) for cell to survive, hence again drugs involving notch pathways being trialed in combo.

More broadly though, and as I said reacently, there is likely to be more ways to terminate a cancer and methods may simply suggest themselves: Stopping tumour cell from proliferating (interfering with Notch pathways) , or cells diffense mechanism being triggered fighting it (efti way), etc.

As such, there may well be many keys to many doors that when opened may help with one or more aspects.

Once again, different cancers do different things. This may well mean that we may need to deal with different pathways (immunotherapy approaches) to treat them all. There may be a grand unified theory but we first need to find our way through separate aspects (for those who know physics, u cannot know how all forces combine into one, unless u know how each one works and then begin to link them together)

From investment point of view, I would find it hard to imagine one can say one therapy is THE winner. Each may (hopefully be the key to one or more aspects). So, in short, i go with where trials are at and IMM is in a good place ... all in my personal opinion.

Comming to fetch data for Phase II in this game is quite a feat!
