What is the advantage of natural macrocycles?

"The clinical successes of macrocyclic natural products [like the fungal-derived cyclosporine] provide tantalizing hints of what could be achieved by the systematic exploration of this compound class."

What if Phylogica could harness natural products and create large-scale synthetic libraries? Perhaps this is exactly what they have been able to achieve. From the Annual Report for 2013;

"[Efforts include the generation of] genetically encoded macrocyclic phylomer libraries that promise to significantly improve the drug-like properties of hits that emerge from the various screens."
