Reasonably, focus is presently on Rett's as major treatment indication for trofinetide.  TBI is a much larger patient population and PII results in severe TBI were encouraging, particularly given an absence of alternative specific treatments.  The following article, just published, raises concern for longterm cognitive deficits (development of dementia) in those with only 'mild' TBI.   I'm sure companies are taking note of any developments for the treatment of TBI as awareness in the public and scientific/medical communities grows.....

Even Mild Traumatic Brain Injury Might Confer Increased Dementia Risk in Veterans

Traumatic brain injury (TBI) — even mild TBI without loss of consciousness — is associated with increased risk for new-onset dementia in veterans, according to a retrospective cohort study in JAMA Neurology.
Using healthcare records, researchers matched nearly 180,000 U.S. veterans diagnosed with TBI between 2001 and 2014 with 180,000 veterans without TBI. During roughly 4 years' follow-up, dementia was diagnosed in 6% of the TBI group versus 3% of the comparison group.
After adjustment for age and other confounders, dementia risk was 2.4-fold higher in those with mild TBI without loss of consciousness compared to those without TBI. Dementia risk increased with the severity of TBI, so that those with moderate-to-severe TBI carried roughly a 3.8-fold increased risk relative to those without TBI.
Editorialists note, "The young mean age of veterans in this study (50 years) raises concerns that this problem will increase as TBI-exposed veterans age."
