While its true I am going, I'm not leaving HC...

I'm going next March to the Pre-eminent Global OA conference!!

Tonight, lets just see what's on the high level agenda, what it means to us and how YOU, yes YOU, are going to be represented!

In March 2023 OARSI are conducting their annual Pre-eminent World congress in Denver, Colorado.
OARSI stands for Osteo Arthritis Research Society International

    Magnificent.

    https://hotcopper.com.au/data/attachments/4791/4791630-467e100f703c0617ef7b4978624671fe.jpg

But Mozz, what's on the agenda and is any of it of interest to us?

Tonight, lets just take a look at the topics and my thoughts on each one.

    Please now enjoy.

OARSI 2023 PROGRAM - HIGH LEVEL

The below are the individual program highlights, but unlike just a parrot I don't just copy and paste them here for your reading wonderment. Mozz research should not be boring, crusty and a rehash of Google searches that anyone can do. Nope, it must be dynamic, on the forefront and certainly associated with YOUR holding.

I have commented on each one to show you WHY it is relevant, WHAT I aim to get out of it...and wait till the last few paragraphs right at the bottom of this post to SEE how YOU will benefit from all of this.

    https://hotcopper.com.au/data/attachments/4790/4790530-d39fb67ca58438847fdb61f38d7c3f26.jpg

Mate, Paradigm released some fairly awesome data in NGF and how pain receptors pick up on this secretion and translate it to pain signals via the nervous system. Can you imagine how insightful such a session might be? Let me at em'. I mean just let there be a question session at the end of this session, my first question will be to the panel..."How would you rate a drug that can down regulate NGF in a safe manner?".

Can you just imagine how good this one starting session is going to be?

    https://hotcopper.com.au/data/attachments/4790/4790531-c410ccd973182d9ba025e1e5ce49c9a9.jpg

Hmm this could be a good one too, what does PAR have as competition into the future? Is there anything that looks promising that's on the cutting edge? How many years ahead are we? I can only do so many personal Google searches, what are the world's best emerging therapies? What do I need to make a note of to study and compare with iPPS?

    https://hotcopper.com.au/data/attachments/4790/4790534-6a40ef98265820e9608acb9ebc35bc8b.jpg

Now this I wanna see...what are the highest rated submitted abstracts...what's popular and why? In terms of worldwide OA research, who's who in terms of forefront research?

    https://hotcopper.com.au/data/attachments/4790/4790539-e4714aea2e5320a93c0d32122a63a64d.jpg

Now I'm taking a bit of a stab here, but by this topic, I get a sneaking suspicion that they are eluding to the fact that the progression of OA in the general populating is not merely attributable to us living longer. Nah, there is more going on and I suspect it has something to do with BMI. 3

A simple suggestion is to modify one's diet and exercise regimes, but like I posted not long ago, this isn't going to be easily implemented. My simple anecdotal evidence is only based on a couple of doctor's evidence, I'm sure this is going to be much more apparent in the case of USA doctor's experiences. The general patient there usually wants a pill, some treatment, yes diet and exercise we all know...but a lot of them often just want a pill/injection/medicine.

    https://hotcopper.com.au/data/attachments/4790/4790542-151471936ff6c14ff56347d38c68ae58.jpg

Well when I read this topic of discussion I think of Tanezumab, I think Fasinumab...NGF complete blockers...perhaps some good results for a slice of the OA population but the side effects were too great including, but not limited to, rapid progressive OA 5, the one thing it should stave off! ...Indeed a pivot is sorely required, that may just be around the corner...

    Hint Hint.

Gee, not a bad first half day....there's more? You bet...read on for Day 2:

    https://hotcopper.com.au/data/attachments/4790/4790543-c2f8b8467632c76cff7495fb91b2cfbf.jpg

Ahhhh decisions decisions, which one to go to? Session 1 or 2? Session 1, how can we prevent OA? I have word of a possible solution...is it too early to bring it up here?

    https://hotcopper.com.au/data/attachments/4790/4790552-1eee7743b637933742812ea1df79ea44.jpg

There have been some studies in regards to calcification that I have come across in the past but they mainly involve older folk and not necessarily related to OA per se.
There is some cutting edge research being done in the area of proteoglycans, their function is diverse from hydrodynamic properties through to providing tensile strength of the connective fibres.6

Yeah Mozz sure, good to read this cutting edge theory but does it relate to us? Why do you get so excited about text book stuff, cant we have a practical example, something WE are involved with?

    So so glad you asked:

"Furthermore, we reveal that the severe RRV-induced joint pathology, including thinning of articular cartilage and loss of proteoglycans in the cartilage matrix, was diminished with treatment. PPS is a promising new therapy for alphavirus-induced arthritis, acting to preserve the cartilage matrix, which is damaged during alphavirus infection. Overall, the data demonstrate the potential of glycotherapeutics as a new class of treatment for infectious arthritis".

(My emphasis added, the above was from Lara J. Herrero et al. See reference 6.5 below)

Prob a fairly complex session, novel therapeutics and Smart Biomaterials, we need to be across this. Its possibly years away if there is proven efficacy.

    https://hotcopper.com.au/data/attachments/4790/4790559-9adcb69158c5ba9e86e9086807477625.jpg

Hmm decision time again. Biomechanics and load and how it pertains to KOA progression would be a good one. I have read in a few articles and papers that misalignment (whether it be natural or surgery invoked) can have a noticeable affect on the progression of OA over time. It accounts for a lot of OA prevalence.

In fact a paper by the infamous Dr David Felson stated that Valgus Malalignment (A mis-alignment from midline) increased lateral risk of OA progression.6.7

New investors to PAR reading this? Dr D Felson is not only an world renowned expert in OA but he is both an MD and a PhD.

Dr. Felson has an AB degree (cum laude) from Harvard College, an MD from Johns Hopkins Medical School and an MPH from Boston University. He completed his clinical training at Case Western University Hospitals (medicine) and Boston University (rheumatology)

Association with OARSI? Who Dr Felson? Are you kidding?

"He has also received the OARSI Lifetime Achievement Award 2012 due to his devotion to increasing the understanding of osteoarthritis". 6.8

    And guess what?

He is a consultant to us. Lovely.

He was instrumental in our excellent Trial design submitted successfully to the FDA. He in fact used to work on the other side of the table...yes he was employed by the FDA to interview Sponsors!

Papers? 268 published papers and counting. This is what PR, Dr Skerett and now Marco and team have organised for us.

    How many investors know this?

    https://hotcopper.com.au/data/attachments/4790/4790561-00f58e70b09d82ff3735e052df73368a.jpg

Adipose tissue is the tissue fat that's just under your skin (subcutaneous fat), but its also present between organs. Indeed in obesity cases there are higher levels of Adipose inflammation. There is an important link here between OA and this layer of Adipose. Don't forget, Adipose also exists in the bone cavities (known also as Bone Marrow Adipose tissue).7

Another split session, imaging would be good to try and get a sense of what they look for and any improvements in this area. I wait patiently for 6 month data release in terms of our super 008 program where hopefully we will start to get a profound sense of how our iPPS can make structural changes, 6 month read isn't a long time away so if we see some decent improvements here, well its going to be exciting.

Hmm more good ramifications for us, Heart Failure with Preserved Ejection Fraction as just one example? New guys - this is the latest pre clinical model we have in place in the background. Its reasons like this I hold a core share parcel. For the super long term.

    https://hotcopper.com.au/data/attachments/4790/4790565-90076a55604b2ed675a94d3c483d8c51.jpg

Sunday starts with a bang...The stats are that after injury, within 10 years you have a 50% chance of acquiring OA.

    https://hotcopper.com.au/data/attachments/4790/4790567-303b3f4bce983fa9e6c40cff757fca0b.jpg

Hmm again another good session, lets see what is out there and how they compare to our own solution?

    https://hotcopper.com.au/data/attachments/4790/4790569-aece4c7957110f51d888eb234e56303e.jpg

Ok I might have to do some serious Biology 101 (maybe its 201) before attending this session, the study of 'Omics' involves multi disciplines such as:
proteomics, transcriptomics, genomics, metabolomics, lipidomics, and epigenomics. 8 *blah*.

I think I might have to carry a dictionary into this one.

    https://hotcopper.com.au/data/attachments/4790/4790572-1e330b7ebc59be334514b222b87c45af.jpg

Ok if I manage to decipher/survive the previous session I might attend this one...otherwise It's concurrent session 9 for me.

    https://hotcopper.com.au/data/attachments/4790/4790573-18af1616415dfeb36c954ecf1a967cee.jpg

Id like to attend this one for sure...over the years I've gotten a real sense of how this should be the first line of attack in terms of OA onset and in fact this is a question I asked my GP buddy the other day. Indeed most of us know we SHOULD exercise more, we SHOULD eat better...we SHOULD employ holistic wellness techniques, but no matter the benefits, how many of us actually do? According to anecdotal evidence from my GP childhood pal, not a lot do...circa 1% !!!

    https://hotcopper.com.au/data/attachments/4790/4790575-e8e49166eab920781996a395373513ef.jpg

Another interesting session, again, will be nice to hear this debate also with what we know about iPPS and just what sort of a 'Breakthrough' it actually is!

    https://hotcopper.com.au/data/attachments/4790/4790577-927a281df6d34739ddf6165531219034.jpg

I do hope by now you guys are getting a real sense of what sort of value this conference is going to be? Reading these topics is sending shivers down my spine (no pun actually intended). I really am not sure if I'm going to be able to keep up with all the incredible material and learnings I'm going to experience at this top notch 4 day seminar. Think Mozz is going to be in some sorta OA Research Nirvana...just wait till I start smoozing with the crowd afterwards. After Party at the OA clinic anyone?

    https://hotcopper.com.au/data/attachments/4792/4792261-57f148721ea05869be02199a1b8a9154.jpg
Mozz Mozz where for art thou? I'm at OARSI 2023

A small example before we go 'beyond other joints and onto the spine'.

At the 2019 AGM I distinctly remember PR commenting about how it was looking like the joints of the hand were the best responding joints to the iPPS treatment. It will really be a session like this that can highlight the pathologies involved and indeed garner some sort of idea of the scope that we have in the important area of the spine that affects so many!

@adherent ? I know you for one would garner a lot out of this one.

One of the best reads I had in terms of PAR announcements was the MOA announcement that dealt with NGF. This was terribly insightful and started a whole research chain for me. To be honest, I still have so much to learn. New to us and are interested in the MOA specifically in the realm of pain and WHY we are so good at reducing it in an OA setting?

A small snippet that's highly relevant to this OARSI session:

Back in 2019 PAR's CSO, Dr Krishnan sought to ask two questions:

    1. Do the bone cells (osteocytes) under the cartilage produce the pain mediator NGF?
    2. Can the drug pentosan polysulfate sodium (PPS) reduce the production of NGF by
    those bone cells?

The answer? A resounding YES and it was peer reviewed.

A small section of the great announcement:

"The implications of the finding of pain reduction via inhibition of NGF secretion in
osteocytes are far-reaching for the multiple actions of PPS relevant for the treatment of OA
complementing its actions in the reduction of inflammation and cartilage protection."

It was this post amongst a few that charged me to add some decent parcels of shares to my initial holdings. (Not advice).
Read reference 9 in the reference section at the very end of this post. This was a most insightful PAR announcement out of the many we have had.

This OARSI session will be a thrill for me, to be able to hear from Global experts of pain and OA and have in the back of my mind the research that we have seen on how we address it, how effective we are and what exactly is the potential for us!

    Amazing with a capital A

The final sessions involve a review of the year, again some interesting and relevant topics, how about the one on Biomarkers (See below)? Again, this session alone will be pretty incredible and highly topical. I wonder if I will meet Dr D Felson and Dr V Kraus? It will be great to hear the presenter's thoughts on biomarkers and map them to what we are observing and the ones we have reported on, to date.

GLAD YOU ARE GOING MOZZ BUT.....

Look Mozz, I'm kinda happy for you that you are going and yeah, think you might learn a thing or two, but how does that help me and my investiture in the said PAR ??

Guys, what's my point in telling you all the above...I'm not being passive..I'm being active.

What do I mean? Yeah I'm going to this conference...it will kill a number of days of annual leave, it will cost some cash that I could've invested directly in you know which company's shares...but I think it is worth it. So worth it.

Not only I'm going, but I'm going with another vested PAR investor.

Why is it important that two of us go???

Because there are concurrent sessions...so when I simply CANNOT decide which session to attend as both sessions are just so darned good, I can go to one, and my fellow PAR holder can go to the other, at night, we compare notes over a nice bottle of red...Don't worry, I will shout him the red on your behalf..

And guess what? Who gets a nice executive summary of the conference and what exactly transpired from OUR perspective?
The need to know points, the important competitors to be wary of, the latest science, Mozz notes ® and of course, the who's who in terms of who was there...

I'm a mere mortal but by gee,
    ...by jingo and
    .........by crickey...

...if I strike a convo up with any Big Pharmas reps or Rheumatologists or Orthopaedic surgeons, I'm going to ask for their opinions and also ask them what they would rate a solution such as ours. Have they heard about iPPS?

Unofficial ambassador to the company and your personal HC PAR rep?

That's me. I'm a vested holder, I want our word to spread. My views are aligned with the maximum prosperity of our company...BUT I am long term....I know what our company is doing....you'll get the short term share price bumps, but these aren't the days I hold out for...its the exciting long term that I wait for.

Yes I can't say how quickly I can get the posts out to you just after each day transpires but I'll be taking lots of notes which I will eventually post!

I will do this primarily for my own benefit but YOU, dear reader, will also benefit.

    YOU just watch this space.

    My Personal views of course

    Mozz
