Great read Tony. It almost makes me laugh everytime i read sections like below. Again with the explanation how hard it is to escape the endosome, and various groups saying how clever Feldan is for finding a way that sort of works. Makes me wonder again why these groups are not aware that apparently we could do this at will a year or more ago. Seems we are just not getting the information out to those who need to know, at least up until now.
Hopefully that is about to change and this report again emphasizes how major our progress could be regarded with correct data to support targeted cell penetration, clear and demonstrated endosomal escape, and high percentage delivery of active ingredients.

"The Feldan shuttle is inspired by an old trick: fuse a positively charged, naturally-occurring molecule called a cell-penetrating peptide (CPP) to proteins to help them cross cell membranes. But so far, CPPs haven’t proven very useful for drug delivery because they tend to get trapped in lipid membrane structures called endosomes as" they enter cells. If the CPP-protein pair can’t escape the endosome and enter the cytosol—the cell’s main compartment—the scheme is useless."
