I agree, I believe the BEN series of CPPs use a number of different mechanisms of action too. And remember, Phylogica have only analysed a handful of the peptides. How many more are being analysed/identified with the second generation libraries and platform enhancements to identify endosomal escape? This is what has me excited more than anything else.
After screening Phylomer® libraries containing billions of different peptides, we identified a panel of unique hits that were successfully able to enter cells. Importantly, teams at Roche and Phylogica independently confirmed several peptides, as having cell selective uptake in cells that line the blood-brain barrier known as 'brain endothelial cells'.
I've also been giving a lot more thought to why Phylogica are focusing on Phenotypic Screening. The new presentation offers many answers which I will come back to later.
