I have taken a look at AOH1996. Any new target to inhibit cancer growth is welcome in human-kinds battle with cancer.

In cancer research there are literally hundreds of targets that are being studied as potential new ways to treat cancer. Specific protein and DNA targets are usually best “targeted” with either small molecule drugs or with monoclonal antibodies, or a combination of the two, as in the case of antibody-drug conjugates. These therapies are very specific in their design so they can “hit” or “inhibit” only a single piece of the cancers armoury. Resistance becomes a problem and cancer escape is a common way patients progress after being on targeted therapy. AOH1996 is an interesting drug in that it inhibits a “cancer hub” where multiple targets rely on that “hub” to be available. Hence the analogy that like an airline, if you knock out a “hub airport” you can cripple an airlines operations.

CF33 is different. Being a potent oncolytic virus it has been designed to specifically infect and grow only in cancer cells. Not only does it grow and burst cancer cells, it releases the mutated insides of cancer cells for recognition by the immune system when this happens. Hence a major way CF33 works, and continues to work, is by immune system clearance of remaining cancer cells, such as specially hard to find metastases.

Could CF33 and AOH1996 be complimentary? Maybe but this would be many years down the track and assumes AOH1996 makes it to market. We are already combining CF33 with a marketed monoclonal antibody KEYTRUDA (a $23Bil per year blockbuster sold by Merck).

A few observations about AOH1996, that in no way diminish the good work published by the COH team.

AOH1996 only inhibits some cancer cells lines. CF33 kills all 60 NCI cancer cell lines outright.
The in vivo data is promising, but CF33 works better in the same models.
