I agree that Starpharmas DEP technology appears to offer superior quality linkage options to currently available ADC technology and products. So, as in your little test, LINKAGE is definitely a key word to emphasise. DEP technology offers 'designer' linkages depending the desired effect ( strong, weak, slow release).

There are three more key words and phrases, describing advantages of DEP ADCs over existing ADCs I would like to add:

PAYLOAD:

DEP ADCs offer far superior payload, with multiple linkages per dendrimer molecule/ nanoparticle that can be 'armed' with a drug or radio pharmaceutical ( or radio diagnostic) agent compared to current ADC technology. The antibody then only needs the one link on its surface to link it to the dendrimer. From my reading, current ADCs have limited ( a small number) of linkages on the antibody protein, to which the active agent is bound.

LESS COMPLEX DEVELOPMENT AND MANUFACTURE:

From my reading, current ADCs are very complex to develop and manufacture, due the difficulty of making those linkages of the active agent to the particular antibody protein complex structure. Each new antibody has a different structure and presents a new complex problem to solve. Starpharma's DEP dendrimers have been linked to many active agents now and it is my understanding that the process is now well understood and it is relatively straightforward to ' reproduce' the process to link new agents. In other words, DEP is a PLATFORM technology. A reproducible process for linking dendrimers to different agents and able to be manufactured cost effectively and at scale. A case study demonstrating the versatility and reproducabilty of the DEP platform technology is how quickly Starpharma developed DEP remdesivir.

DELIVERY: Superior solubility and ' designer' size of starpharma's dendrimers improves delivery to where they are most needed ( more targeted), improving efficacy and reducing toxicity. (As impressively seen with DEP docetaxol in both the preclinical studies and then in the case study in the current phase 2 trial, where there was around 60 times greater concentration in the tumour biopsy than surrounding tissue - BETTER EFFICACY through more targeted delivery. Also seen in the phase two trials was less hair loss and bone marrow toxicity - LESS TOXICITY due also to more targeted delivery. There is no reason why these advantages should not also apply to DEP ADCs.

I agree, a great product. But the proof will be in the marketing. Especially up against (inferior) local competing products that have more local market knowledge and networks. Great products don't sell themselves.
