Bisantrene is an old compound, but you are the one behind the times.
Bisantrene's primary MoA is the inhibition of FTO (126x lower IC50 than topoisomerase II).
Dysregulated FTO expression (m6A levels) is implicated in oncolytic viral therapy resistance.
Essentially, little old bisantrene targets the system that is preventing your drug from working in patients...Oncolytic viruses aren't special either, because dysregulated FTO (m6A levels) is implicated in chemoresistance across 25 different drug classes. Bisantrene has overcome antimetabolite resistance in patients achieving 57% CR rate and 40% ORR in patients refractory to last line of treatment.
